Why Callus is not developing?
i am using usual onion root tips to grow callus. i have added all macro & micro nutrients, vitamins to the medium and 1, 2 & 3% auxins and cytokinens. 16hrs light per day for 7 days, but still no callus development. i am not getting the problem? any solutions?
It looks like you were doing everything right. Micropropagation can be a very finicky process. The success, even under ideal circumstances, is very low. If you did not have many replications (~10 per petri with several petris), it is possible that you won't see successful callus formation.
The other possibility is that the root stock you are using for propagation is old. Old, travel-damaged cells mean no propagation--ie samples from the grocery store tend to do poorly and require more replications than usual to form a callus.
